Overview

Antioxidant molecule determination is the identification and quantification of specific compounds responsible for a sample's antioxidant capacity, distinguishing the contributions of individual constituents from bulk measures of total activity. Whereas global assays report aggregate radical-scavenging power, molecular determination resolves which species, such as phenolic acids, flavonoids, carotenoids, ascorbate, tocopherols, thiols, or polysaccharide-bound antioxidants, drive the observed effect. This typically combines separation and detection techniques, including chromatographic profiling, with mechanism-specific radical-scavenging and reducing assays, allowing structure-activity relationships to be inferred from the substitution patterns and redox potentials of the isolated molecules. The subject matter examined in this area includes the isolation of antioxidant flavonoids from aerial plant parts, correlation of total phenolic content with measured capacity in medicinal plants, evaluation of micronutrient antioxidants, and characterization of radical-scavenging behavior in cell-free and cardiomyoblast systems and in isolated mitochondria subjected to oxidative stress.
